       郑静博士具有分子生物学、肿瘤生物学、生物化学和神经生物学等多方面的技术背景和研究经验,负责并主持过多项神经系统疾病和肿瘤领域的项目申请与执行,如上海市浦江计划、科技部863项目、国自然青年基金、上海市自然基金等。已经在CarcinogenesisJournal of Neuroscience, Journal of Biological Chemistry, Neurobiology of Disease, J Neurooncol.NeuroscienceBrain Research BulletinAJTR等学术刊物上发表论文二十余篇,被SCI他人引用近百余次。研究方向如下所述:

 

  

l  神经系统智力障碍疾病发生发展的分子机制与药物靶标开发     

  

 智力发育障碍又称为精神发育不全或弱智,是儿童期最常见的神经系统疾病之一。引起智力障碍的原因可以是遗传性的,如多基因或单基因突变;也可以是非遗传性的,如神经系统感染,药物滥用,或脑损伤。其主要症状是智力发育迟缓,适应能力低下,注意力不集中等。部分病人(20-30%)伴随有癫痫发作。智力障碍的发病率约为2-3% 。在欧洲,约有8% 的医疗费用于智力疾病的治疗,远远高于其他同类疾病的费用。因此,智力障碍疾病成为医学领域中最重要的有待解决的难题之一。在小鼠中选择性的在特定脑区敲除与智力障碍疾病相关的基因,通过分析突触可塑性、学习记忆和癫痫发生等指标研究这些基因的功能;小鼠特定脑区中在单个神经元水平敲除与智力障碍疾病相关的基因,并且使该神经元可视化,从而研究这些基因在神经元形态学、发育和可塑性中的作用。上述工作陆续发表在20152011年度的Neuroscience 2014年度的Brain Research Bulletin 2013年度的PNAS2008年度的J. Biol. Chem.2010年的Neurobiology of Disease2011年的Int J Clin Exp Path上。 
 

  

l  神经系统肿瘤发生发展的分子机制与药物靶标开发      

  

胶质瘤是一种常见的脑肿瘤,发生于神经外胚层。按细胞种类分,胶质瘤的主要类型有星形胶质细胞瘤,少突胶质细胞瘤和胶质母细胞瘤。2007年,世界卫生组织按胶质瘤的恶性程度和预后的好坏情况将胶质瘤分为IIIIIIIV,四个等级。I级胶质瘤通常是良性的,在儿童中发病更频繁,一般不会发展为更高级别肿瘤,其基因变化也与更高级别的胶质瘤不同,预后良好。II级属分化良好性,虽然不是良性的,但预后较好,中位生存期为5-8年。III级是渐变性星形胶质细胞瘤,中位生存期为3年。IV级的胶质母细胞瘤,恶性最强,中位生存期为12-18个月,大于60岁患者的生存期低于中位生存期。我们利用CRISPR/Cas9基因编辑技术、RNA干扰技术和基因过表达/点突变技术等,结合肿瘤细胞生长曲线、克隆形成、细胞周期、细胞凋亡、细胞迁移、血管生成、裸鼠皮下成瘤和原位成瘤等检测技术,研究我们关注的药靶基因对神经系统肿瘤发生和发展的影响,以期望寻找到药物作用的靶标,开发基于基因治疗和药物治疗的新产品。上述工作陆续发表在2015年度的AJTR2014年度的CarcinogenesisInt J Clin Exp Path上,2013年度的J Neurooncol.2012年度的J Am Chem Soc.2010年的J. Neurosci.20112015年度的ACS Nano上。
